การคัดแยกแบคทีเรียที่ผลิตเอนไซม์อะไมเลสทนร้อนจากแหล่งดิน

บทคัดย่อ
งานวิจัยนี้มีจุดประสงค์เพื่อคัดเลือกแบคทีเรียที่ผลิตอะไมเลสทนร้อนจากดิน โดยเก็บตัวอย่างดินจากบริเวณ พระนาคปรกเมืองไพร อาคารคณะศิลปศาสตร์และวิทยาศาสตร์ และทางเข้ามหาวิทยาลัยราชภัฏร้อยเอ็ด นำตัวอย่างดินแหล่งละ 1 กรัม มาผสมในอาหาร NB นำไปบ่มที่อุณหภูมิที่ 37 องศาเซลเซียส เป็นเวลา 24 ชั่วโมง และบ่มต่อที่อุณหภูมิที่ 60 องศาเซลเซียส เป็นเวลา 48 ชั่วโมง ทำการคัดกรองแบคทีเรียที่สามารถผลิตอะไมเลสได้โดยนำแบคทีเรียไปเลี้ยงบนอาหาร NA ที่ผสมแป้ง 1 เปอร์เซ็นต์และทดสอบกิจกรรมของอะไมเลสด้วยสารละลายไอโอดีน ผลการคัดแยกพบแบคทีเรียทนร้อนที่ผลิตอะไมเลส 3 ตัวอย่าง คือ AN01 AN02 และ AN03 นำแบคทีเรียที่คัดแยกได้ไปทำสอบกิจกรรมของอะไมเลสที่อุณหภูมิ 50 60 70 80 90 และ 100 องศาเซลเซียสโดยใช้น้ำแป้ง (pH 7.0) เป็นสารตั้งต้น ผลการศึกษาพบแบคทีเรีย AN02 มีกิจกรรม อะไมเลสสูงสุดที่ 60 องศาเซลเซียส (0.183 ยูนิตต่อมิลลิลิตร) จากผลการจัดจำแนกแบคทีเรียด้วยลำดับของดีเอนเอของยีน 16SrRNA ด้วยฐานข้อมูล EzBioCloud พบว่าแบคทีเรีย AN02 คือ Bacillus aryabhattai ที่ค่าความเหมือนที่ 99.5%
