ฤทธิ์ต้านออกซิเดชัน และฤทธิ์ความเป็นพิษต่อเซลล์มะเร็งของรากมะข่วง

บทคัดย่อ

สารสกัดรากมะข่วงเตรียมได้โดยการสกัดรากแห้งของมะข่วง (Zanthoxylum rhetsa) ด้วยเทคนิคการแช่หมักในตัวทำละลายเฮกเซน เอทิลแอซิเตต และเมทานอล ได้สารสกัดหยาบชั้นเฮกเซน เอทิลแอซิเตต และสารสกัดชั้นเมทานอล ตามลำดับ สารสกัดทุกชนิดใช้ในการศึกษาปริมาณฟีนอลิกและปริมาณฟลาโวนอยด์ทั้งหมด ฤทธิ์ต้านออกซิเดชัน และฤทธิ์ความเป็นพิษต่อเซลล์มะเร็ง นอกจากนี้ยังมีการศึกษาสารพฤกษเคมีเบื้องต้นเพื่อหาสารเมแทบอไลต์ทุติยภูมิกลุ่มหลัก การทดสอบด้วยรีเอเจนต์ที่เหมาะสมพบว่าสารสกัดทุกชนิดมีกลุ่มสารเมแทบอไลต์ทุติยภูมิหลากหลายชนิด โดยเฉพาะอย่างยิ่งสารแอลคาลอยด์ซึ่งเป็นสารกลุ่มหลัก สารสกัดชั้นเมทานอลมีปริมาณฟีนอลิกทั้งหมดและปริมาณฟลาโวนอยด์ทั้งหมดมากที่สุด รองลงมาคือสารสกัดชั้นเอทิลแอซิเตตและสารสกัดชั้นเฮกเซน สารสกัดชั้นเมทานอลแสดงฤทธิ์การกำจัดอนุมูลอิสระ DPPH (IC50; 1.12 ± 0.01 mg/mL) และความสามารถในการรีดิวซ์ได้สูงที่สุดเมื่อเทียบกับสารสกัดชนิดอื่นๆ จากผลการทดสอบฤทธิ์ต้านออกซิเดชันของสารสกัดพบว่ามีความสัมพันธ์เชิงบวกกับปริมาณฟีนอลิกและปริมาณฟลาโวนอยด์ทั้งหมด ผลของการทดสอบฤทธิ์ความเป็นพิษต่อเซลล์พบว่าสารสกัดชั้นเฮกเซน เอทิลแอซิเตต และสารสกัดชั้นเมทานอลแสดงฤทธิ์ความเป็นพิษต่อเซลล์มะเร็งเฉพาะมะเร็งปอดชนิดเซลล์เล็ก NCI-H187 อย่างจำเพาะเจาะจงและมีประสิทธิภาพด้วยค่า IC50 เท่ากับ 22.78, 26.50 and 3.37 μg/mL ตามลำดับ งานวิจัยนี้แสดงให้เห็นว่าสารสกัดของรากมะข่วงเป็นแหล่งใหม่ของสารต้านอนุมูลอิสระจากธรรมชาติ เพื่อการปกป้องหรือแม้แต่การป้องกันโรคภัยที่เป็นอันตรายต่อชีวิต  นอกจากนี้รากของมะข่วงอาจเป็นอีกทางเลือกที่มีศักยภาพเพียงพอต่อการพัฒนาสู่การรักษาโรคมะเร็งที่มีความจำเพาะเจาะจงสูง
