Effects of Phosphate Nutrients on the Growth of the Marine Microalga Navicula sp. Isolated from the Eastern Coast of Thailand

Navicula sp. เป็นไดอะตอมทะเลที่มีบทบาทสำคัญในระบบนิเวศทางน้ำและมีศักยภาพในการใช้ประโยชน์ในอุตสาหกรรมเพาะเลี้ยงสัตว์น้ำและเทคโนโลยีชีวภาพ งานวิจัยนี้ศึกษาผลของความเข้มข้นฟอสเฟตที่แตกต่างกันต่อการเจริญเติบโตและการสังเคราะห์แสงของ Navicula sp. TISTR 11108 ที่แยกจากชายฝั่งตะวันออกของประเทศไทย โดยเพาะเลี้ยงในอาหาร F/2 ที่มีฟอสเฟตแตกต่างกัน 3 ระดับ ได้แก่ 21 µM (ชุดควบคุม), 10.5 µM และ 5.25 µM ติดตามการเจริญเติบโตด้วยการวัดความหนาแน่นเซลล์และปริมาณคลอโรฟิลล์ เอ เป็นเวลา 14 วัน พบว่าชุดควบคุมมีการเจริญเติบโตและการสังเคราะห์แสงสูงสุด โดยมีความหนาแน่นเซลล์ ~94 ×104 เซลล์/มล. คลอโรฟิลล์ เอ ~16 มก./มล. และมีอัตราการเจริญจำเพาะที่ 0.28 ต่อวัน ส่วนชุดฟอสเฟตต่ำมีการเจริญเติบโตและการสะสม คลอโรฟิลล์ เอ ต่ำกว่า และเซลล์เสื่อมสภาพเร็วขึ้น การย้อมไนล์เรดเพื่อตรวจสอบหยดน้ำมันที่มีีอยู่ในเซลล์สาหร่ายพบว่าทุกการทดสอบมีการสร้างหยดน้ำมัน โดยชุดการทดสอบฟอสเฟตที่ความเข้มข้น 10.5 µM มีหยดน้ำมันมากที่สุด รองลงมาคือ 5.25 µM และ 21 µM ตามลำดับ ผลการศึกษาชี้ว่าฟอสเฟตเป็นธาตุอาหารหลักที่ส่งผลต่อชีวมวล ประสิทธิภาพการสังเคราะห์แสงและการผลิตน้ำมันของ Navicula sp. TISTR 11108 ดังนั้นการควบคุมระดับฟอสเฟตจึงเป็นปัจจัยสำคัญในการพัฒนาเทคโนโลยีการเพาะเลี้ยงสาหร่ายอย่างยั่งยืน
