In vitro Anticoagulant Effect of Red Rice Bran Extract on Human Blood

Abstract

Coagulation abnormalities are key features observed in various diseases. This study aimed to evaluate the anticoagulant effect of red glutinous rice bran extract (Oryza sativa L.) on blood coagulation in vitro, using plasma from healthy volunteers. The extract was prepared at concentrations of 5, 10, and 20 mg/mL, corresponding to final concentrations in the test tubes of 1, 2, and 4 mg/mL, respectively. Blood coagulation was assessed using Prothrombin time (PT) and Activated partial thromboplastin time (aPTT) assays. The results showed that at a concentration of 1 mg/mL, the PT and aPTT values were 26.53 ± 0.27 and 121.23 ± 1.07 seconds; at 2 mg/mL, PT was 39.77 ± 0.36 seconds and aPTT was 164.20 ± 0.47 seconds; and at 4 mg/mL, PT and aPTT were 67.30 ± 0.64 and 174.90 ± 0.49 seconds, respectively. Glutinous rice bran extract at concentrations of 10 and 20 mg/mL (final concentrations of 2 and 4 mg/mL in vitro) significantly prolonged the prothrombin time (PT) compared to the PBS control group (negative control), with a P-value < 0.05. A comparison between the groups treated with 5 mg/mL and 20 mg/mL extract (final concentrations of 1 and 4 mg/mL in vitro) showed a statistically significant difference in activated partial thromboplastin time (aPTT) results (P < 0.05). These findings indicate that increasing the concentration of red rice bran extract prolonged coagulation time, with 4 mg/mL showing the strongest anticoagulant activity. This study suggests that Oryza sativa L. extract may have potential as an anticoagulant agent and could be further explored for therapeutic or preventive applications in coagulation-related disorders.
